- The antagonist will be The Second of the Fallen.
- It is speculated that Keanu Reeves will reprise his role as John.
- With that being said, it has also been rumoured that Paul Bettany will portray Constantine.
- The film will provide insight on John's childhood, including him learning how to use his abilities and even his first exorcism.

- DC Comics announced a sequel to the 2005 Constantine movie was in the works, with producer Lorenzo di Bonaventura linked to the project. He stated: "I'd love to do it... We want to do a hard, R-rated version of it. We're going to scale back the size of the movie to try and persuade the studio to go ahead and make a tough version of it."
- In September 2010, Warner Brothers commissioned a sequel script from Frank Cappello, one of the original Constantine's two credited writers. The story is said to be an original idea from Cappello and does not draw from any previous published Hellblazer stories. The screenplay's tag line is "Through the Eye of The Needle".
- In November 2012 it was announced that Guillermo del Toro had signed on as writer and director for a Justice League Dark film centered around DC Comics' supernatural characters, with John Constantine featuring as a main character.
- By May 2019, Reeves stated that he is open to reprising the role in the future.
- By July 2020, it was reported that a new film centered around the titular character was in development with J.J. Abrams serving as producer. The project is said to be a joint-production between DC Films, Warner Bros. Pictures, and Bad Robot Productions. The film is said to be similar in tone the filmmaker's other upcoming DC projects, including an HBO Max exclusive Justice League Dark television series, as well as a film centered around Zatanna Zatara.
- In November 2020, Stormare announced on an Instagram post that a sequel was "in the works." Representatives for Warner Brothers and Reeves did not immediately respond to a request to comment.
